Mechanism of Action

Boswellic acids --- principally AKBA --- are potent selective inhibitors of 5-lipoxygenase (5-LOX), the enzyme responsible for leukotriene (LTB4) synthesis. This mechanism is distinct from NSAIDs (which target COX-1 / COX-2) and therefore carries a gastric-protective profile, making Boswellia suitable for patients who cannot tolerate conventional anti-inflammatory agents. Additional mechanisms include NF-κB suppression, TNF-α / IL-1β / IL-6 downregulation, NLRP3-inflammasome pathway inhibition, and MMP-3 / MMP-9 inhibition --- the last of which protects cartilage matrix from proteolytic degradation in osteoarthritis.
